Precisely engineering 3-D brain tissues

ScienceDaily (Nov. 29, 2012) ? Borrowing from microfabrication techniques used in the semiconductor industry, MIT and Harvard Medical School (HMS) engineers have developed a simple and inexpensive way to create three-dimensional brain tissues in a lab dish.

The new technique yields tissue constructs that closely mimic the cellular composition of those in the living brain, allowing scientists to study how neurons form connections and to predict how cells from individual patients might respond to different drugs. The work also paves the way for developing bioengineered implants to replace damaged tissue for organ systems, according to the researchers.

"We think that by bringing this kind of control and manipulation into neurobiology, we can investigate many different directions," says Utkan Demirci, an assistant professor in the Harvard-MIT Division of Health Sciences and Technology (HST).

Demirci and Ed Boyden, associate professor of biological engineering and brain and cognitive sciences at MIT's Media Lab and McGovern Institute, are senior authors of a paper describing the new technique, which appears in the Nov. 27 online edition of the journal Advanced Materials. The paper's lead author is Umut Gurkan, a postdoc at HST, Harvard Medical School and Brigham and Women's Hospital.

'Unique challenges'

Although researchers have had some success growing artificial tissues such as liver or kidney, "the brain presents some unique challenges," Boyden says. "One of the challenges is the incredible spatial heterogeneity. There are so many kinds of cells, and they have such intricate wiring."

Brain tissue includes many types of neurons, including inhibitory and excitatory neurons, as well as supportive cells such as glial cells. All of these cells occur at specific ratios and in specific locations.

To mimic this architectural complexity in their engineered tissues, the researchers embedded a mixture of brain cells taken from the primary cortex of rats into sheets of hydrogel. They also included components of the extracellular matrix, which provides structural support and helps regulate cell behavior.

Those sheets were then stacked in layers, which can be sealed together using light to crosslink hydrogels. By covering layers of gels with plastic photomasks of varying shapes, the researchers could control how much of the gel was exposed to light, thus controlling the 3-D shape of the multilayer tissue construct.

This type of photolithography is also used to build integrated circuits onto semiconductors -- a process that requires a photomask aligner machine, which costs tens of thousands of dollars. However, the team developed a much less expensive way to assemble tissues using masks made from sheets of plastic, similar to overhead transparencies, held in place with alignment pins.

The tissue cubes can be made with a precision of 10 microns, comparable to the size of a single cell body. At the other end of the spectrum, the researchers are aiming to create a cubic millimeter of brain tissue with 100,000 cells and 900 million connections.

Answering fundamental questions

Because the tissues include a diverse repertoire of brain cells, occurring in the same ratios as they do in natural brain tissue, they could be used to study how neurons form the connections that allow them to communicate with each other.

"In the short term, there's a lot of fundamental questions you can answer about how cells interact with each other and respond to environmental cues," Boyden says.

As a first step, the researchers used these tissue constructs to study how a neuron's environment might constrain its growth. To do this, they placed single neurons in gel cubes of different sizes, then measured the cells' neurites, long extensions that neurons use to communicate with other cells. It turns out that under these conditions, neurons get "claustrophobic," Demirci says. "In small gels, they don't necessarily send out as long neurites as they would in a five-times-larger gel."

In the long term, the researchers hope to gain a better understanding of how to design tissue implants that could be used to replace damaged tissue in patients. Much research has been done in this area, but it has been difficult to figure out whether the new tissues are correctly wiring up with existing tissue and exchanging the right kinds of information.

Another long-term goal is using the tissues for personalized medicine. One day, doctors may be able to take cells from a patient with a neurological disorder and transform them into induced pluripotent stem cells, then induce these constructs to grow into neurons in a lab dish. By exposing these tissues to many possible drugs, "you might be able to figure out if a drug would benefit that person without having to spend years giving them lots of different drugs," Boyden says.

German producers plan Pope Benedict biopic

MUNICH, Germany (Reuters) - Two German producers have bought the film rights to an upcoming biography of Pope Benedict by the Bavarian author of three best-selling interview books with the pontiff.

The Odeon Film company said producers Marcus Mende and Peter Weckert planned a film for international release based on a biography by journalist Peter Seewald due to be published in early 2014.

Seewald's book-length interviews with Benedict - two as Cardinal Joseph Ratzinger and one as pope - have given readers many insights into the life and thoughts of the shy theologian who now heads the Roman Catholic Church.

Seewald has signed on as a consultant to the scriptwriter, Odeon Film said in a statement on Thursday. It gave no information about the schedule for the film or who might play the main role.

"The producers plan an international film that illustrates all aspects of the extraordinary life and work of Joseph Ratzinger from his birth on Easter night in 1927 in Marktl am Inn in Bavaria to his pontificate today," it said.

Benedict's predecessor Pope John Paul was the subject of a dozen documentary films around the world and two major television movies in the United States.

Oil rises on higher economic growth expectation

NEW YORK (AP) ? The price of oil rose Thursday on an improved reading on U.S economic growth while lawmakers continued their tussle over the "fiscal cliff."

The Commerce Department Thursday raised its estimate for U.S. economic growth to an annual rate of 2.7 percent in the summer quarter of this year. That's much higher than the 2 percent rate estimated a month ago and more than twice the 1.3 percent rate in the three previous months.

Also Thursday The National Association of Realtors said its seasonally adjusted pending home sales index rose in October to its highest level in nearly six years.

Benchmark crude rose $1.58 to close at $88.07.

In other trading, Brent crude, which is used to price international varieties of oil, rose $1.25 to $110.76 per barrel on the ICE Futures Exchange in London.

In a wild Silicon Valley Bowl shoot-out at Foothill College in Los Altos Hills, the Delta College Mustangs scored 63 points against the DeAnza Dons, eking out a narrow 63-56 victory over the 5th ranked NorCal Conference co-champion.

This is the second game in a row the Mustangs (8-3) have scored 63 points against DeAnza (8-3) in a bowl game. Last year, they routed an injury riddled Dons team 63-13 in Togo?s Delta Bowl, played at Delta?s DeRicco Stadium. This year, DeAnza?s high powered offense had all its star players ready to rumble, relishing a chance for revenge. ?Key among those was the NorCal Conference offensive player of the year, quarterback Josh McCain.

Spearheaded by Delta quarterback Sam Hutsell?s balanced passing attack, the rushing of De?Marieya Nelson, receiver Jack Killian and two key defensive stops in the first quarter, Delta burst out to a 28-7 lead. It didn?t take long for DeAnza quarterback Josh McCain to start reeling the Mustangs back in. The DeAnza Don?s own balanced attack, combined with McCain?s scrambles and the rushing of Alfonso Brister, matched Delta?s 21 point second quarter.? By halftime, Delta still controlled the game with a commanding 34-21 lead.

As you might expect, the scoring was far from over.? Sam Hutsell?s one yard touchdown run and Jack Killian?s 37 yard TD reception added 14 more points to Delta?s total in the third quarter.? Not to be outdone, DeAnza added 21 points of its own with two McCain touchdown passes and a five yard touchdown run by Brister. By the end of the quarter, Delta still clung to a 49-42 lead.

DeAnza took their first lead of the game with 9:48 left in the fourth quarter.? McCain turned another scramble into a 30 yard touchdown run to go up 56-49. Delta worked its way back into a tie with a drive-finishing ten yard touchdown run by De?Marieya Nelson. ?A huge defensive stop ended DeAnza?s next drive. Defensive end Elijah Saina finally sacked McCain to set up a failed 3rd down pass attempt by the DeAnza quarterback, forcing a punt.

Time was short?2:36 remained in the game. The Mustangs needed to drive 88 yards for the victory. Utilizing a balanced attack, Delta efficiently drove 75 yards. With 10 seconds left, De?Marieya Nelson, who was battling a lingering hamstring injury all game, was called to duty. Nelson bulled his way the final 13 yards?extending the ball over the goal line to give the Mustangs a nail-biting 63-56 victory! Both teams combined for almost 1,200 yards in offense.

Coach Gary Barlow was ecstatic after the white-knuckle last second victory, but as a coach, was still frustrated at seeing 56 points scored against the Mustang defense. ?All that matters is?if you score one more point than your opponent, you?ll be happy at the end!? This was Coach Barlow?s first Silicon Valley Bowl victory, and 10th bowl appearance in 12 seasons at Delta. The Mustangs have been to seven straight bowl games, with this win marking Delta?s first back-to-back bowl championships.

DeAnza quarterback Josh McCain was named the ?Offensive Player of the Game? throwing for 347 yards on 26 of 45 attempts with four touchdowns. Delta receiver Jack Killian was the game?s MVP, catching eight passes for 172 yards and two touchdowns.

'Hot air' may spike climate talks

The right of some rich countries to keep using "hot air" carbon permits could hamper progress at UN climate talks starting on Monday in Qatar.

With the Kyoto Protocol running out at the end of the year, several countries want to carry over unused carbon allowances.

Campaigners say this "hot air" could render new carbon cuts meaningless.

And there are fears that old fault lines between rich and poor will prevent any significant developments.

This year's meeting comes just a month before the expiration of the first commitment period of the Kyoto Protocol, the world's most significant global agreement on reducing emissions of carbon dioxide.

Last year in South Africa, negotiators agreed what is called the Durban Platform. The nub of this was a trade off - the richer countries would extend their carbon cutting beyond 2013 via an extension of the Kyoto Protocol. In return, the developing nations would negotiate a new deal by 2015 that would commit all countries, including emerging super-powers like India and China, to reduce emissions from 2020.

Key to that small forward step was a "coalition of the willing" - an unlikely alliance of the European Union, small island states and the least developed countries.

'Difficult issue'

But in Doha, some of the fine details of an agreement have to be worked out. And these are proving very troublesome for that group in particular.

One of the concerns is over "hot air" - several EU countries were given huge allowances of carbon permits that they want to carry over into a new commitment period. But the scale of the surplus, some 13 billion tonnes of carbon dioxide, could render future promises to cut carbon effectively meaningless.

"Many (delegates) are unhappy about the carry-over," said Dr Quamrul Chowdhury, a negotiator from Bangladesh who speaks for the developing countries group. "It could be a difficult issue."

Tracy Carty, a climate change policy adviser from Oxfam agrees that the issue of surplus carbon permits could be very important in the negotiations.

"Otherwise we aren't going to get the emissions reductions that we need."

Another critical element is money. During the COP15 talks in Copenhagen in 2009, richer countries agreed to provide $30bn in fast-start finance for the period up to 2012. They also established a Green Climate Fund to control up $100bn per year expected to be raised by 2020 to help poorer countries mitigate the effects of climate change.

Empty fund

But several countries including the UK, France and Germany have been accused of reclassifying money that was already committed as development aid. According to research carried out by Oxfam, only 33% of this climate finance can be considered as new.

And with no money in the climate fund after the end of this year, Oxfam believes it is the most critical issue at this meeting.

Tracy Carty argues the dispute could hamper the new alliances that have led to progress at previous meetings in Cancun and Durban.

"On the broad elements of 2015, we aren't going to see any 'big-bang' moments in Doha like we saw in Durban. But on finance it is a crossroads - this is a period where we are coming to the end of a tranche of money and we don't know what's on the table for next year," she said.

Some, though, remain hopeful that Doha can see a "big-bang" moment, pointing to the fact that the new leadership in China and the re-elected President Obama are both making optimistic noises about climate.

"President Obama indicated that he would make some progress on global warming," said Dr Chowdhury, "and the new leadership of China might go for a deal - that is my personal point of view but we want to see that progress."

LAUSD English Learners To Move Faster Into Mainstream Classes

Triggered by a federal civil rights investigation, Los Angeles Unified has launched a network of programs designed to more quickly move English learners into mainstream academic classes and help close the achievement gap.

The strategies are detailed in the 150-page English-Learner Master Plan, which the district overhauled last year after a U.S. Department of Education probe determined that English learners and African-American students were being denied access to educational opportunities.

Through more rigorous instruction and increased support, officials say that English learners and students who struggle with standard English can acquire the language skills needed to tackle math, social studies, science and other courses required for graduation.

"Let me paint a picture," said Hilda Maldonado, director of LAUSD's Multilingual and Multicultural Education Department, which oversees its language-development programs.

"There's a kid with a backpack that holds the math and social studies and all of the other lessons they need to learn. There's also an English learner who has two backpacks -- one with all of the academics, the other that contains the skills of learning a new language.

"This is the duality of the challenges these kids face," Maldonado said. "The content and language compete at the same level."

The nation's second-largest school system, Los Angeles Unified has more English learners than any other district -- about one-third of its 600,000 students. Spanish is spoken by 94 percent of English learners, with dozens of other languages spoken by small percentages of youngsters.

The district's programs for English learners mandate daily language-development instruction -- 45 minutes to an hour for elementary students and one to three periods for secondary students.

Within those parameters, there are programs to accommodate the range of student experiences and the levels of proficiency.

There is remedial instruction for newcomers for instance, and support programs for students with more advanced skills. Dual-immersion programs teach language fluency and academic proficiency in both English and the target language for students who want a cross-cultural education.

Under the district's previous bilingual education programs, 38 percent of English learners tested early advanced or better in their English skills in 2011-12. At the same time, about 15 percent of English learners were reclassified as proficient, opening the door to more academic options.

However, nearly 40 percent of English learners failed to achieve proficiency after five years of instruction. Getting these kids reclassified as proficient is a key goal of the master plan.

And even after students are deemed proficient, the district will be tracking their academic progress to ensure they remain on track.

"Reclassification isn't the end of your career -- go forth and speak English," Maldonado said. "We're going to be monitoring them and stepping in if we see there are problems."

The new programs come at a critical time for English learners in Los Angeles Unified, which has ratcheted up its graduation requirements so that all high school students must pass a slate of college-prep courses in order to get a diploma.

Additionally, a more rigorous curriculum known as the Common Core will take effect in 2014, impacting lessons at every grade level.

"I think the rigor of the master plan is the most important element," said school board member Nury Martinez, who recalled struggling as an English learner during her childhood in Pacoima.

"We want to make these kids feel capable of learning English and not segregating them," she said. "It has nothing to do with their intelligence or their ability to retain the information. It just takes a little longer with us."

Puerto Rico a backdoor into US mainland for drugs

A vicious drug war has come to the island home of 4 million Americans, which is being used as a transshipment point to the East Coast. NBC's Gabe Gutierrez reports.

SAN JUAN, Puerto Rico ? The raids begin before dawn. Fierce knocks followed by shouts of ?Polic?a!? rattle neighborhoods while a helicopter hovers overhead.

On this day in late September, heavily armed agents swarm otherwise peaceful-looking homes throughout the island.

In Toa Alta, outside of San Juan, one suspected drug trafficker surrenders without incident. Amazingly, no neighbors wander outside to see what?s going on. They must already know ? or would prefer not to.

By 8 a.m., 16 people are in handcuffs, facing charges that include drug trafficking and money laundering.


The arrests are part of "Operation Overtime." U.S. Immigration and Customs Enforcement's Homeland Security Investigations special agents, working with officers from the Puerto Rico Police Department and San Juan Police Department, launched the initiative to combat drug trafficking in the Caribbean.

This is a war on American soil.

The murder rate in Puerto Rico is higher than in any U.S. state ? and it?s roughly six times the national average. The island ? best known for its stunning beaches and rich history ? is home to roughly 4 million American citizens.

The island?s police superintendent, Hector Pesquera, told NBC News that more than two-thirds of the homicides are tied to the drug trade.

Luis Romero Font, 60, and his wife Marie Jane Rodriguez, 57, know the pain that comes with the incessant violence. Their son, Julian, was murdered during a robbery in April 2011.

He had just celebrated his 20th birthday.

Luis Romero Font and Marie Jane Rodriguez remember their son, who was killed last year in San Juan. Puerto Rico's murder rate is now roughly six times the U.S. national average.

"I try to focus my thoughts away from the negative, away from what I don?t have, and I try to remember what I did have,? his mother told NBC News. "That?s what sustains me."

'A hero had died'
Julian was stabbed to death as he walked down the street with his girlfriend. The robber cut him three times as Julian lunged to protect her.

"A hero had died," his father said, choking back tears. "I am very proud of him. I am very saddened ? [by] this huge weight I have to continue bearing."

The teenager who killed Julian made off with his cell phone ? a valuable tool that is often stolen for drug traffickers to communicate with each other.

In memory of his son, Romero Font, a telecommunications company CEO, has become an activist, starting his own group called "?Basta Ya!" meaning, "Enough is Enough!"

"Puerto Rico cannot survive with this horrendous crime wave," he said.

Violent crime has been a part of life on the island for decades ? ever since the 1980s when Colombian cartels began to thrive here.

But the last several years have seen a dramatic spike.

According to the Puerto Rico Police Department, there were 1,016 murders in 2010. That number rose to 1,136 in 2011.

Outgoing Gov. Luis Fortuno credits a police crackdown for reducing homicides in certain parts of the island by 22 percent so far this year. But he stresses it?s not enough.

He told NBC News that he feels ignored by the federal government and that he?s "banging on Washington?s door" for more help.

Federal agents and local police are cracking down on drug trafficking in Puerto Rico. NBC News speaks with an ICE agent and the island's recently appointed police superintendent, and tags along on a raid.

"We are American citizens," Fortuno said. "And we deserve to be protected."

Puerto Rico?s governor-elect, Alejandro Javier Garcia Padilla, was not immediately available for comment.

Earlier this summer, federal agents arrested dozens of workers at Puerto Rico?s main airport who had allegedly helped smugglers move cocaine to the mainland.

Drug seizures rising
According to ICE, agents have seized 22,000 pounds of illegal drugs so far this year. That?s up from 13,961 pounds in 2011 and 9,275 in 2010.

The street value of the recovered cocaine alone totals $250 million, ICE said.

Since Puerto Rico is a U.S. territory, once illegal drugs reach the island, they?re unlikely to face further U.S. customs inspections on their way to the mainland.

"I think the cartels are smart," said Rep. Michael McCaul, R-Texas. "They realize we're putting all of our efforts on the Southwest border (with Mexico) and we're not paying attention to the 'third border' ? the Caribbean ? so now they're exploiting that."

In June, McCaul chaired a hearing on Capitol Hill that focused on the increased drug smuggling in Puerto Rico.

"We have no strategy for the Caribbean," McCaul told NBC News. "There is no comprehensive plan."

He said that hopefully would change quickly as Washington learns more about the problem. In July, Homeland Security Secretary Janet Napolitano visited the island and promised to study the issue, adding that an increase in aid to Mexico did not come at the expense of the Caribbean.

A senior homeland security official told NBC News that since then, the department had conducted a comprehensive review of the situation in Puerto Rico and planned to focus on "quality over quantity."

The official, who asked not to be identified, said any future manpower surge would likely be "modest" and the department?s priority would be to increase efficiency among the officers already on the ground as well as resources outside the island ? instead of simply pouring more money into the operations.

The increased cooperation would help prevent the flow of illegal weapons to the island as well as illegal drugs from the island, the official said.

Police department 'broken'
The drug-trafficking crackdown comes as Puerto Rican police fight to reclaim the trust of residents.

The 17,000-member force is the second largest in the United States. In a report last year, the Justice Department accused officers of engaging "in a pattern and practice of misconduct that violates the Constitution and federal law."

According to the DOJ?s findings, the department was "broken in a number of critical ways," including repeated instances of civil rights violations and illegal searches and seizures.

In 2010, the FBI arrested dozens of law enforcement officers on the island for allegedly providing protection to drug dealers.

Earlier this year, Hector Pesquera, the former head of the FBI's Miami division, was named superintendent of the Puerto Rico police. He told NBC News that weeding out corruption within his department has been a priority ? and he?s seen progress.

"We have streamlined our procedures to work on the internal affairs cases,? Pesquera said. "We're getting rid of the backlog that existed. The word out there is clear: 'If you commit a transgression, you're going to be terminated.'"

For now, Pesquera said he and other Puerto Rican leaders are simply asking for a shift in federal resources. He said he realizes the difficult fiscal position the U.S. is in, but "we should not be begging for assistance."

Luis Romero Font, the activist who lost his son, agrees. He?s pushing the U.S. government to help stop the violence.

"When you lose a son, it?s like losing a huge part of yourself," he said. "Either [the U.S.] stops this now, or this crime wave will become something that infects Florida and the rest of the U.S. down the line."

So away from the tourists, the raids continue. America?s "third border" ? the place known as the "island of enchantment" ? remains at war.

Calif. city building a "tsunami-resistant" port

(AP) ? It doesn't matter if the earth sways in Chile, Alaska or Japan, the formation of the sea floor along the U.S. West Coast generally aims any tsunami surges at the tiny California port town of Crescent City. Churning water rushes into the boat basin and then rushes out, lifting docks off their pilings, tearing boats loose and leaving the city's main economic engine looking as if it has been bombed.

That's what happened in March 2011, when a Japanese earthquake sparked a tsunami that sank 11 boats, damaged 47 others and destroyed two-thirds of the harbor's docks.

Port officials are hoping that tsunami is among the last of many that have forced major repairs in Crescent City, a tiny commercial fishing village on California's rugged northern coast. Officials are spending $54 million to build the West Coast's first harbor able to withstand the kind of tsunami expected to hit once every 50 years ? the same kind that hit in 2011, when the highest surge in the boat basin measured 8.1 feet and currents were estimated at 22 feet per second.

Officials are building 244 new steel pilings that will be 30 inches in diameter and 70 feet long. Thirty feet or more will be sunk into bedrock. The dock nearest the entrance will be 16 feet long and 8 feet deep to dampen incoming waves. The pilings will extend 18 feet above the water so that surges 7 ? feet up and 7 ? feet down will not rip docks loose.

Crescent City was not the only West Coast port slammed by the tsunami, which was generated by a magnitude-9.0 earthquake in Japan. The waves ripped apart docks and sank boats in Santa Cruz, Calif., and did similar damage in Brookings, Ore., just north of Crescent City. But their geographical location doesn't make them as vulnerable to multiple tsunamis.

"Normally, Crescent City takes the hit for all of us," said Brookings harbormaster Ted Fitzgerald.

Since a tidal gauge was installed in the boat basin in 1934, this small port has been hit by 34 tsunamis, large and small. It typically suffers the most damage and the highest waves on the West Coast, said Lori Dengler, professor of geology at Humboldt State University.

The sea floor funnels surges into the mouth of Crescent City's harbor, and the harbor's configuration magnifies them, experts say.

A wave generated by an earthquake in Alaska on Good Friday, 1964, killed 11 people and wiped out 29 city blocks. That was 10 years before the boat basin was even built.

When the waves hit in 2011, the port was still repairing damages from a tsunami that hit in 2006. Officials already had a plan for dealing with future tsunamis, said Ward Stover, owner of Stover Engineering in Crescent City, which put together the plan.

With no tsunami building codes, Stover said the state of California and Crescent City decided to prepare for the kind of tsunami expected to hit every 50 years. They rejected as too expensive building a tidal gate to close off the mouth of the harbor or trying to survive a powerful tsunami like the one that hit in 1964. Instead, they planned to make the docks strong enough to ride out the most likely surges.

"It's tsunami-resistant, not tsunami-proof," Stover said.

Construction has been marked by one delay after another. Government funding was slow, and a custom-built drill bit for installing the extra-strength pilings deep in bedrock broke. So authorities switched to installing temporary docks the old-fashioned way, by pounding in the pilings, to get them through the winter. Many of the 60 commercial fishing boats based in Crescent City are still mooring in the outer harbor. Others have to make do without water or electricity.

The March 2011 tsunami was a wake-up call for communities up and down the West Coast. Many improved tsunami evacuation plans and held mock evacuations.

But some experts say the West Coast is still not taking the threat seriously enough.

"Many ports on the West Coast are in denial as to their tsunami hazard," said Costas Synolakis, professor of civil and environmental engineering and director of the Tsunami Research Center at the University of Southern California.
